New cholesterol reagent.

چکیده
R ECENTLY a new reagent was proposed for the determination of cholesterol (1, 2). This reagent was purported to be saturated ferrous sulfate in glacial acetic acid. Since it has been reported that ferrous sulfate is qualitatively insoluble in anhydrous acetic acid (3, 4) and that ferrous iron is “subject to pronounced air oxidation in glacial acetic acid” (5), it was felt that several factors should be investigated concerning the reagent. These factors are as follows: 1. The solubility of ferrous sulfate in glacial acetic acid under the described conditions (2) 2. The stability of this ferrous ion in the same system 3. The solubility of ferric sulfate in glacial acetic acid under identical conditions, since this compound is suspected of being the active agent in this reaction 4. The reaction spectra for cholesterol with both ferrous and ferric sulfate solutions

Ferric chloride spray detector for cholesterol and cholesteryl esters on thin-layer chromatograms.
The use of a stable solution of ferric chloride for the detection of cholesterol and cholesteryl esters on thin-layer chromatographic layers is described. Its advantages over the commonly used Carr-Price reagent are listed, and the levels of sensitivity for the new reagent are shown. Detection of other lipid classes is also possible.
متن کاملReagent for the enzymatic determination of serum total cholesterol with improved lipolytic efficiency.
We describe a sensitive method for quantifying the extent of cholesterol ester cleavage during enzymatic assay of total cholesterol in serum. Lipids are extracted from the assay mixture with chloroform/methanol (1/1 by vol), concentrated, then quantified by "high-performance" thin-layer chromatography. Although with conventional enzymatic reagents for determination of serum total cholesterol th...
متن کاملInfluence of incomplete cholesteryl ester hydrolysis on enzymic measurements of cholesterol.
Esterolytic activity of three enzymic cholesterol reagents was evaluated with use of human serum-based reference materials. Primary cholesterol standards were used to calibrate the enzymic methods, and incomplete cholesteryl ester hydrolysis was measured as the bias between the enzymic cholesterol values and corresponding reference values. The average biases observed with eight reference specim...
متن کاملImproved method for determination of high-density-lipoprotein cholesterol II. Enzymic determination of cholesterol in high-density lipoprotein fractions with a sensitive reagent.
A reagent is described for the colorimetric enzymic determination of high-density-lipoprotein (HDL) cholesterol. The reagent can be used with HDL fractions isolated by the various methods of precipitation of low- and very-low-density lipoproteins we investigated. The considerable sensitivity obtained by use of Barham-Trinder's reaction allows the sample/reagent volume ratio to be decreased to 1...
متن کاملMethod for fully automated determination of total cholesterol in blood serum, including saponification and extraction.
A fully automated method has been developed for the determination of total cholesterol in blood serum, using the AutoAnalyzer system. According to the new method, based on the cholesterol determination of Abell et a!. (1), the serum sample is saponified and subsequently extracted with carbon tetrachloride. After treatment with Liebermann-Burchard reagent, the amount of cholesterol is determined...
